Vertebrate neuron-specific nuclear protein or NeuN (Neuronal Nuclei) reacts with most neuronal cell types throughout the nervous system including cerebellum, cerebral cortex, hippocampus, thalamus, spinal cord and neurons in the peripheral nervous system including dorsal root ganglia, sympathetic chain ganglia and enteric ganglia. NeuN is primarily localized in the nucleus of the neurons with light staining in the cytoplasm by immunohistochemical staining. There are few cell types, including Purkinje, mitral and photoreceptor cells, that are not reactive with NeuN antibody. NeuN is developmentally expressed and first observed by immunoreactivity assay shortly after neurons have become postmitotic. No staining is observed in proliferative zones. NeuN is an excellent marker with a molecular weight rang from 46 kDa to 48 kDa for neurons in primary cultures and in retinoic acid-stimulated P19 cells. It is also useful for identifying neurons in transplants.
