[Active ingredient]

Artecom is compoud preparation, one tablet contains 32mg Dihydroartemisinin, 320mg Piperaquine Phosphate and 90mg Trimethoprim.

[Character]

Artecom is a green coated tablet, it is off-white or light yellow removing the coating.

[Therapeutic indication]

Artecom is a compound antimalarial drug recommended for the treatment of uncomplicated Plasmodium falciparum and Plasmodium vivax malaria, in particular, in the case of resistance to other antimalarials.

[Dosage]

Oral administration, For adults, take 2 tablets for the first time, followed by 2 tablets at the 6th, 24th and 48th hour.The total dose is 8 tablets. For children, the dosage should be decreased accordingly. Dosage adjustment is as shown in the table below:

[Adverse reactions]

1.Digestive tract: nausea, vomiting, impaired appetite, abdominal pain, diarrhea and salivation.

2.Nerve system: dizziness, headache, tinnitus, asthenia, hyposomnia.

3.Allergic reaction: skin itch and eruption

4.Laboratory abnormal: transient peripheral erythropenia, transient increase of ALT and AST, leukopenia and thrombocytopenia.

[Contraindications]

1.Those who are allergic to any of the three active ingredients are prohibited to use.

[Pharmacological and toxicological actions]

Pharmacological actions

Artecom is a compound antimalarial drug containing Dihydroartemisinin, Piperaquine phosphate and Trimethoprim. Dihydroartemisinin has strong gametocidal action against plasmodium against body, and can kill plasmodium rapidly, controlling the symptoms immediately. Piperaquine phosphate is 4-aminoquinolines antimalarials, The antimalarial action of Piperaquine phosphate is similar with chloroquine, but it is non-cross resistant. After oral administration, Piperaquine phosphate is absorbed and is stored in liver, then it is slowly released into blood, with a long-lasting action and is therefore used for pevention and treatment against the symptoms of malaria. Trimethoprim has the function of inhibiting dihydrofolte reductase. It combines with the dihydrofolte reductase of plasmodium interfering with the synthesis of purine so as to inhibit the growth of plasmodium. Pharmacological studies show that the combination of the three ingredients has synergistic effect, and postpone drug resistance of plasmodium.

Toxicological study

Dihydroartenisinin:

Genotoxicity: The reslut of Ames Test, CHL Chromosome aberration test and micronucleus Test show negative.

Reproductive toxicity: have a embryo-fetal toxicity for pregnancy mice, increase absorption of fetal and show dose-dependency. No tertogenic action.

Piperaquine phosphate: 

Repeated dose toxicity test: dose one time for a week in dogs, 100mg/kg for 14 weeks or 25mg/kg for 26 weeks, the reslut is that target prgan toxicity is liver.

Genotoxicity: The reslut of Ames Test, Chromosome of bone marrow cell analysis and sister chromatid exchange test show negative.

Reproductive toxicity: No tertogenic action and mbryo-fetal toxicity in animal experiment.

[Pharmacokinetics]

After oral administration, Artecom is absorbed by gastrointestinal tract, where dihydroartemisinin is well adsorbed with rapid effect, wide distribution and rapid excretion and metabolism. In 30 minutes after oral administration, piperaquine phosphate is detected, while trimethoprim is rapidly and completely absorbed. Blood peak concentration is reached in 1-2 hours. After absorption, Artercom is mainly distributed in liver, kidney, intestine, blood and body fluids. It is mainly excreted via kidney and intestinal tract.
